Metastasis and angiogenesis in cancer are dependent on a variety of biologically active compounds including serine protease. Inhibition of proteases such as plasmin is a potential route for suppression of both metastasis and angiogenesis. Synthesis of tetrahydrothiopyran-4-one based inhibitors containing either a dialkylamino chain, a lysine analog side chain and a dipeptide (D-ile-phe or D-ile-Trp), or just the D-ile-phe dipeptide was accomplished. A compound with both the lysine analog side chain and the dipeptide but lacking a carbonyl was also produced.
