Stable isotopes (2H, 13C) are useful for studies on the metabolism of endogenous or exogenous compounds. GC/MS makes it possible to measure partial loss of 2H by exchange from polydeute-rated molecules and to detect compartmentation of precursors. Oxidoreduction can be studied by measurements of the loss of isotope from the carbon carrying the hydroxyl group. Label in the coenzyme can be corrected for by the use of mixtures of differently labelled compounds. Isotope effects can be used to study mechanisms in vitro and pathways in vivo. Labelling in vivo with 2H-substrates makes it possible to distinguish coenzyme pools. Topics discussed in this symposia include: Synthesis and Applications of Organic Compounds Labelled With Isotopic Hydrogen; Applications of Stable Isotopes and NMR Spectroscopy for In Vivo Studies; Radiation-Induced Reactions; Applications of Isotopically-Labelled Compounds in Clinical Research; Isotopes in Organic and Bio-Organic Reaction Mechanisms; New Techniques in Isotopic Analysis; Synthesis and Application of Organic Compounds Labelled With Iodine Isotopes; Synthesis of Isotopically-Labelled Compounds Via Organometallic Chemistry; Synthesis and Applications of Organic Compounds Labelled With Isotopic Carbon, Sulphur and Other Isotopes; The Use of Isotopically Labelled Compounds in Studies of Drug Metabolism, Disposition and Toxicity; Synthesis and Applications of Labelled Compounds in Positron Emission Trans-Axial Tomography; Applications of Radioactively-Labelled Compounds to In Vitro and In Vivo Research.(AW).

Compounds labeled with carbon-14 and tritium are indispensable tools for research in biomedical sciences, discovery and development of pharmaceuticals and agrochemicals. Preparation of Compounds Labeled with Tritium and Carbon-14 is a comprehensive, authoritative and up-to-date discussion of the strategies, synthetic approaches, reactions techniques, and resources for the preparation of compounds labeled with either of these isotopes. A large number of examples are presented for the use of isotopic sources and building blocks in the preparation of labeled target compounds, illustrating the range of possibilities for embedding isotopic labels in selected moieties of complex structures. Topics include: Formulation of synthetic strategies for preparing labeled compounds Isotope exchange methods and synthetic alternatives for preparing tritiated compounds In-depth discussion of carbon-14 building blocks and their utility in synthesis Preparation of enantiomerically pure isotopically labeled compounds Applications of biotransformations Preparation of Compounds Labeled with Tritium and Carbon-14 is an essential guide to the specialist strategies and tactics used by chemists to prepare compounds tagged with theradioactive atoms carbon-14 and tritium.

Marking the 200th National Meeting of the American Chemical Society, The Division of Nuclear Chemistry and Technology hosted a group of about 90 scientists from 15 different countries to discuss the new trends in radiopharmaceutical synthesis, quality assurance and regulatory control. This event took place in Washington, D.C. on August 27-30, 1990. When I first suggested the idea for this symposium, a group of scientists who pioneered the proposed topics offered their help to organize and run such a big task with me. Their names are listed here in appreciation. Thomas E. Boothe Cyclotron Facility, Mt. Sinai Medical Center, Miami Beach, Florida, USA Robert F. Dannals Division of Nuclear Medicine, The Johns Hopkins Medical Institutions, Baltimore, Maryland, USA Anthony L. Feliu Julich Nuclear Research Center, Julich, Germany Joanna S. Their effort is sincerely acknowledged.

Drug Discovery and Evaluation has become a more and more difficult, expensive and time-consuming process. The effect of a new compound has to be detected by in vitro and in vivo methods of pharmacology. The activity spectrum and the potency compared to existing drugs have to be determined. As these processes can be divided up stepwise we have designed a book series "Drug Discovery and Evaluation" in the form of a recommendation document. The methods to detect drug targets are described in the first volume of this series "Pharmacological Assays" comprising classical methods as well as new technologies. Before going to man, the most suitable compound has to be selected by pharmacokinetic studies and experiments in toxicology. These preclinical methods are described in the second volume „Safety and Pharmacokinetic Assays". Only then are first studies in human beings allowed. Special rules are established for Phase I studies. Clinical pharmacokinetics are performed in parallel with human studies on tolerability and therapeutic effects. Special studies according to various populations and different therapeutic indications are necessary. These items are covered in the third volume: „Methods in Clinical Pharmacology".
