Since the potential toxicity of silver nanoparticles (Ag NPs) has raised serious concerns in the biomaterials and biomedical engineering community, Silver Nanoparticles for Antibacterial Devices: Biocompatibility and Toxicity brings together the synthesis, the physicochemical properties and the biological actions of Ag NPs, as well as the clinical demands for fabricating antibacterial medical devices, discussing how to suppress the side effects of nanomaterials and how to impart to them the selective toxicity. This book presents the two primary paradigms that have emerged in probing the antibacterial applications of Ag NPs, i.e. the active attacking releasing way and the conservative defending approach by taking advantage of various short-range actions; it shows readers how the ways in which Ag NPs have behaved can be engineered purposively. With contributions from leading international experts and extensive references listed in each chapter, this volume provides the general principles on controlling the physicochemical behaviors of nanomaterials and managing their toxicity risks.

Nanostructures for Antimicrobial Therapy discusses the pros and cons of the use of nanostructured materials in the prevention and eradication of infections, highlighting the efficient microbicidal effect of nanoparticles against antibiotic-resistant pathogens and biofilms. Conventional antibiotics are becoming ineffective towards microorganisms due to their widespread and often inappropriate use. As a result, the development of antibiotic resistance in microorganisms is increasingly being reported. New approaches are needed to confront the rising issues related to infectious diseases. The merging of biomaterials, such as chitosan, carrageenan, gelatin, poly (lactic-co-glycolic acid) with nanotechnology provides a promising platform for antimicrobial therapy as it provides a controlled way to target cells and induce the desired response without the adverse effects common to many traditional treatments. Nanoparticles represent one of the most promising therapeutic treatments to the problem caused by infectious micro-organisms resistant to traditional therapies. This volume discusses this promise in detail, and also discusses what challenges the greater use of nanoparticles might pose to medical professionals. The unique physiochemical properties of nanoparticles, combined with their growth inhibitory capacity against microbes has led to the upsurge in the research on nanoparticles as antimicrobials. The importance of bactericidal nanobiomaterials study will likely increase as development of resistant strains of bacteria against most potent antibiotics continues. Shows how nanoantibiotics can be used to more effectively treat disease Discusses the advantages and issues of a variety of different nanoantibiotics, enabling medics to select which best meets their needs Provides a cogent summary of recent developments in this field, allowing readers to quickly familiarize themselves with this topic area

The most common type of adverse events in healthcare in Canada reported by the Canadian Institute for Health Information (CIHI) are nosocomial infections. Amongst nosocomial infections, implant associated infections have been reported to be most common. Despite having the implantation surgeries carefully performed, a small, but still considerable number of devices gets colonized by bacteria resulting in implant associated infections and/or surgical site infections. The patients are then started on high dose antibiotics, which if ineffective, is followed by reimplantation surgeries that leads to long hospital stays and detrimental effects in their lives. Due to this, an alternative to antibiotics is required which could prevent and/or treat bacterial colonization on implants. The main objective of this thesis was to demonstrate the effectiveness of an antimicrobial based CLKRS peptide capped silver nanoparticle coating on a metallic and polymeric based biomaterial used in various implantable medical devices. The CLKRS peptide capped silver nanoparticle formulation was specifically engineered and tested for its antibacterial and antibiofilm properties. Silver nanoparticles were synthesised by photochemical reduction of silver ions upon photocleavage of the photoinitiator I-2959. The metal nanostructure surfaces were protected with the CLKRS peptide and tested on planktonic and biofilms of P. aeruginosa, S. aureus and S. epidermidis. The bacterial quantification was done by survival colony counting. The cytotoxicity of the silver nanoparticle formulation was also tested on human dermal fibroblast, mouse bone marrow derived macrophages, and human epithelial cells by cell proliferation assay. Results show the formation of a nanometric layer of nanosilver on the surface of the material inhibiting the growth of bacteria and eradicating pre-existing biofilms with no significant cell toxicity suggesting the prepared formulation could be a useful tool in preventing and controlling infections on implants during surgery and post implantation. This technology thus could serve as an alternative therapy for surgical site infections and/or implant associated infections.

Nanotechnology is an emerging field of science. It has increased applications in diverse area for the development of new materials at nanoscale levels. Synthesis of nanoparticles using biological methods is referred as greener synthesis of nanoparticles. Green synthesis provides advancement over chemical and physical method as it is cost effective, environment friendly, and safe for human therapeutic use. Stingless bees are highly social (eusocial) insects which populated the tropical earth 65 million years ago longer than honey bees. Among the most common uses of stingless bee honey are to treat stomach disturbance, cough, tonsillitis, sore throat, stomach and intestinal ulcers, cold, disease of the mouth, mucus membrane, and as a wound dressing due to its antimicrobial activity. Stingless bees honey were used to for the green synthesis of silver nanoparticles. Antimicrobial activity of the green synthesised nanoparticles were tested used agar diffusion method against Escherichia coli (E. coli), Pseudomonas aeruginosa, Staphylococcus aureus, Salmonella typhi and Klebsiella pneumoniae. The results showed that stingless bee honey could be effectively used for the synthesis of silver nanoparticle. The synthesized silver nanoparticles shows antibacterial activity on both Gram positive and Gram negative bacteria. This biosynthesis of nanoparticles is cost efficient, pollutant free and simpler to synthesize.

Silver nanoparticles (AgNps) have attracted much interest in biomedical engineering, since they have excellent antimicrobial properties. Silver nanopolymer composites have applications in biochemical sensors, antimicrobial activity and drug delivery system. Silver nanoparticles are more effective than ionic homologues (Ag+) for their antimicrobial activity. Antimicrobial properties of silver nanoparticles are used by their incorporation into medical devices, tissues and other health-related products for skin pathologies to reduce the risk of contamination and to promote higher preventive infection control. Novel hybrid material thin films based on various polymeric systems with embedded silver nanoparticles were synthesized using various methods. The electrical, optical and plasmonic responses of AgNps onto thin layers of polymer composites show encapsulation of nanoparticles. The antibacterial activity of AgNps/polymer composites against various common bacteria is discussed in this chapter. The antibacterial activity of the synthesized hybrid materials was tested against various bacteria, commonly found in hospital environment. Silver nanostructures have especially been of interest because of contrast agents for biomedical image. Shunts used for hydrocephalous silicon elastomer grafted with hydrogel, polyvinylpyrrolidone (PVP) soaked in various antibiotics proved to be active for longer time.
