The past and future of inflammatory pharmacology research: a hot topic in health and disease Inflammation is a physiological response to a traumatic injury, bacterial, or viral infection. However, if not appropriately controlled, it contributes to a long list of diseases, including asthma, atherosclerosis, multiple sclerosis, arthritis, and cancer. Different are the types of inflammatory responses. Acute inflammation is an immediate body response to the cellular damage induced by pathogens, noxious stimuli, or physical injury – it is a short-term response resulting in healing via time-dependent changes of leukocyte functions. First, a leukocytes infiltration happens within the damaged region with the purpose of eliminating the stimulus and repairing the tissue. Chronic inflammation, by contrast, is a prolonged and dysregulated response where the active inflammation contributes both to tissue destruction and to the development of many chronic human conditions and diseases. In the context of exaggerated inflammation, which occurs as a consequence of severe burns or trauma, the body response called sepsis can be associated with fatal outcome. Increased knowledge of the cellular and molecular mechanisms taking part in the different types of inflammation is a central requirement to develop more effective and safer treatments. This is a necessary step to prevent potential severe consequences, i.e., organ failure associated with tissue fibrosis. The mission of Inflammation Pharmacology (section of Frontiers in Pharmacology) is to publish scientifically sound studies that advance our knowledge on different aspects of inflammation and contribute to the development of more effective and safer anti-inflammatory agents. Within the present eBook are collected the top articles published in the Inflammation Pharmacology section in the last 10 years. Some articles explored the roles played by different lipid mediators generated from arachidonic acid, including leukotrienes and prostanoids [such as prostacylin and prostaglandin(PG)F2a], in inflammatory conditions. Moreover, the protectin (PD) family of specialized pro-resolving mediators biosynthesized from the two omega-3 polyunsaturated fatty acids docosahexaenoic acid (DHA) and n–3 docosapentaenoic acid (n–3 DPA) were described for their biological effects, the G-coupled protein receptors pharmacology, biosynthesis, and medicinal chemistry. Some other articles focused on the development of novel strategies to counteract inflammation or to induce its resolution. The current concepts and controversies on classification, pathogenesis, and clinical management of cutaneous adverse events induced by biologic agents used in the treatment of rheumatologic conditions were discussed in another article. The whole-exome and whole-genome sequencing data identifying new and old loci associated with atherosclerosis will lead to discovering new molecular targets for blocking atherosclerosis even in its early stages. This critical issue was reviewed in another paper. Numerous information on an individual clinical condition is held in their platelet-derived microparticles (MPs); the assessment of their number and size together with their content can represent the signature to acquire diagnostic information and to monitor the efficacy of therapeutic agents. Some other articles discussed the role of fibroblasts in the development of fibrosis and potential therapies under investigation. It was enlightened the role of the activation and transdifferentiation of hepatic stellate cells (HSCs) into contractile, matrix-producing myofibroblasts (MFBs) as central events in hepatic fibrogenesis, and summarized the current strategies for targeted delivery of drugs to pro-fibrogenic liver cells, including the development of therapeutics specifically targeting HSCs. The present book is an attempt to describe the most recent developments in the area of pericyte biology which is one of the emergent hot topics in the field of molecular and cellular biology today. Here, we present a selected collection of thirteen detailed chapters on what we know so far about pericytes in distinct organs in physiological and pathological conditions. Further, it provides an update on the most novel functions attributed to these cells and will introduce a newer generation of researchers and scientists to the importance of these cells, ranging from their discovery in different organs through current state-of-the-science. It will be invaluable for both advanced cell biology students as well as researchers in cell biology, stem cells and vascular research. This volume explores pericytes' physiologic roles in different tissues, ranging from the pancreas, lungs and liver through skeletal muscle, gut, retina and more. Together with its companion volumes Pericyte Biology in Disease and Pericyte Biology – Novel Concepts, Pericyte Biology in Different Organs presents a comprehensive update on the latest information and most novel functions attributed to pericytes. To those researchers newer to this area, it will be useful to have the background information on these cells' unique history. It will be invaluable for both advanced cell biology students as well as researchers in cell biology, stem cells and researchers or clinicians involved with specific organs.

Hepatic sinusoidal cells such as sinusoidal endothelial cells, Kupffer cells, hepatic stellate cells, and pit cells play an important role in hemodynamic and metabolic function and are involved in various liver diseases. The frequent involvement of sinusoidal cells in pathologic conditions is explained by the diverse actions of those cells, an understanding of which is essential to clinicians. Generously illustrated, this volume presents results of recent studies on hepatic sinusoidal cells in liver diseases, with an introduction to the structure and function of the various types of sinusoidal cells. The in-depth focus of the book is on the role of hepatic sinusoidal cells in relation to liver injury and regeneration, hepatic sinusoidal microcirculation, alcoholic liver diseases, hepatic fibrosis, liver tumor, liver transplantation, cholestasis, and congenital lipidosis, thus providing a valuable reference source for practitioners and researchers.

Nelson Fausto The Greek myth of Prometheus with its picture of a vulture feasting on its chained victimhas traditionallyprovided a visualimageofliverregeneration. Itis apowerful and frightening representationbut ifone were to substitute the vulture by a surgeon and Prometheus by a patient laying on a properly prepared operating table, the outcomeoftheprocedurewould not differ significantlyfrom that describedbyGreek poets. Yet few of us who work in the field have stopped long enough to ask where this myth originated. Did the poet observe a case of liver regeneration in a human being? Was it brilliant intuition or perhaps, literally, just a 'gut feeling' of a poet looking for good rhymes that led to the prediction that livers grow when part of the tissueisremoved? Thisbookdoesnotattemptto solve these historical issues. Itdoes, instead, cover in detail some of the major modem themes of research on liver regen eration, injury and repair. As indicated in Dr. N. Bucher's chapter, the modem phase ofexperimental studies on liver regeneration started in 1931 with the publication by Higgins and Anderson of a method to perform a two-thirds resection of the liver of a rat. The technique described has 3 remarkable features: 1) it is highly reproducible, resulting in the removal of 68% of the liver, 2) it has minimal if any mortality, and 3) it consists only of blood vessel ligation and does not involve cutting through or wounding hepatic tissue.
