In 1980, the IVth International Cyclic Nucleotide Conference was held in Brussels. As this meeting attracted many investigators involved in cyclic nucleotides and calcium role in intracellular regulation, it was thought that this opportunity could be used to organize, prior to the Congress, an in-depth introductory course on the subject. This was carried out as a NATO Advanced Study Institute. The participants included Ph. D. students and M. D. s engaged in a research training, but also fully trained and well known researchers who wanted a refresher course on the whole subject. During the course, most of the participants and lecturers asked to be provided with a text summarizing the basic lectures of the course. This book was therefore conceived as a basic textbook on the regulation and action of intracellular signal molecules, concentrating mainly on cyclic nucleotides and calcium. It was deliberately kept at a basic level. We would therefore be happy if it could be used as an introduction for interested M. D. s or Ph. D. s working in other fields or entering this field and as a general refresher for researchers interested in the subject. For this reason, very general schemes have been asked of the authors, along with reading lists of available reviews rather than extensive bibliographies. Current Topics in Cellular Regulation: Volume 25 is a collection of papers that deals with GTP, the central regulator of cellular anabolism, and the quantitative approach to metabolic control. , Other papers describe the inactivation and aldolases by limited proteolysis by lysosomal cathepsin M, the regulatory functions of proline, pyrroline-5-carboxylic acid, as well as the characteristics of a hormonal regulation of amino acid transport. One paper suggests that GTP, possibly acting through high guanine nucleotide energy charge, acts to stimulate a wide variety of anabolic processes involved in cell growth or proliferation. Another paper proposes that lysosomes are responsible for the degradation of proteins and other macromolecules, and in regulating cytosolic enzymes. This action is due to the presence of selected proteinases (located on the outer lysosomal surface) that are active at neutral pH, and also which catalyze only limited proteolysis of cytosolic proteins. One paper describes the characteristics and hormonal regulation of a specific transport system for neutral amino acids, namely, the System A-mediated transport. The collection can prove beneficial for biochemists, micro-biologists, bio-physicists, cellular researchers, and academicians involved in the study of cellular biology.

Today, the basic mood of researchers and clinical investigators, both at the center and on the periphery of interferon studies, is optimistic regarding the future of interferons as therapeutic substances. Many also feel these polypeptides will prove invaluable probes in unraveling certain fundamental biochemical processes which control the life cycle and developmental pattern of many human cells. In contrast, only a year or two ago, this optimism had given way to an attitude almost of disenchantment as public and scientific expectations were raised steeply, then rapidly waned, as it turns out, prematurely. Both the mUltiple actions of interferons (a virtual cascade of biochemical reactions may be induced, as documented herein) and the high visibility of interferon research provided by the millions of dollars invested both by national health agencies and by multinational pharmaceutical companies, contributed to an upsweep in public attention to drug development probably unprecedented in this century. Virtually every oncologist, it would seem, was plagued by requests for the experimental agent, although they already had therapies of more proven value. As recently as 1980, even though interferon had achieved success against certain cancers and certain viral diseases, the variability in clinical results was seemingly ever present and little evidence emerged to suggest interferons could cure advanced diseases. Why then the resurgence of an optimistic mood? There are almost always many elements which contribute to happiness, and this is certainly true of the broad frontier of interferon and its place in biochemical research and treatment.
