Mathematical Modeling and Immunology An enormous amount of human effort and economic resources has been directed in this century to the fight against cancer. The purpose, of course, has been to find strategies to overcome this hard, challenging and seemingly endless struggle. We can readily imagine that even greater efforts will be required in the next century. The hope is that ultimately humanity will be successful; success will have been achieved when it is possible to activate and control the immune system in its competition against neoplastic cells. Dealing with the above-mentioned problem requires the fullest pos sible cooperation among scientists working in different fields: biology, im munology, medicine, physics and, we believe, mathematics. Certainly, bi ologists and immunologists will make the greatest contribution to the re search. However, it is now increasingly recognized that mathematics and computer science may well able to make major contributions to such prob lems. We cannot expect mathematicians alone to solve fundamental prob lems in immunology and (in particular) cancer research, but valuable sup port, however modest, can be provided by mathematicians to the research aspirations of biologists and immunologists working in this field.

Interactions between the immune, endocrine and nervous systems seldom appear as main issues in the neurosciences and in immunology. So far this was most likely due to the need to focus on the molecular and cellular bases of single neural, endocrine and immune processes. But hormones, neurotransmitters and neuropeptides can also influence more subtle mechanisms underlying immune cell activity. The contents of this volume aim at listing some aspects which show that not only the bases for neuroendocrine control of more refined mechanisms related to the organization and functioning of the immune systems to exist, but also that the immune system can actively communicate with neuroendocrine structures. The evidence is divided into three categories: - Anatomical, cellular and molecular bases for the exchange of information between immune, endocrine and neural cells, - reciprocal effects between immune and neuroendocrine mechanisms, and - immune-neuroendocrine regulatory circuits. Immunologically triggered neuroendocrine responses can be either beneficial or deleterious for the host. A systematic approach would imply the simultaneous evaluation of neuroendocrine and immune parameters and thus provide the basis for therapeutic interventions based on antagonizing or blocking undesirable effects.

Immunoregulation is one of the areas which has witnessed the most explosive advances of immunology during the past decade. It is in this area that the current view of the immune system has arisen and developed. There is indeed little doubt that immune reactions are primarily determined by messages which are genera ted within the immune system and passed among different types of immunologie cells. This cell communication not only determines the type, intensity and duration of the response after perturbation of the immune system by exogenous antigens, but it is also essential for preventing autoimmune reactions and their clinical conse quences. In order to assure aperfect balance within the enormous com plexity of the immune system, it is not surprising that multiple self-regulatory mechanisms are organized at different levels, such as antibody feedback, idiotypic-anti-idiotypic responses, suppres sor and helper T cells, lymphokine signals and genetic require ments. A nu mb er of observations in recent years have, however, demonstrated that consistent contributions to the immunological homeostasis are given also by signals generated outside of the immune system, namely,in the central and autonomous nervous system as weIl as in the endocrine apparatus. Furthermore, the interactions between the immune system and the other body homestatic mechanisms seem to be bidirectional: if immunological cells may be targets of neuroendocrinological factors, immunological products seem in turn to contribute to the neuro endocrine homeostasis.
