In July 1994, the Canadian Inflammation Society in Toronto hosted a symposium entitled "Novel molecular approaches to anti-inflammatory therapy" as a satellite of the xnth International Congress of Pharmacology. For several reasons the above topic was singled out and separated from the general Congress. Major advances have recently been made in the understanding of inflammatory mechanisms. These advances serve as the basis for a new generation of therapeutic agents which may potentially alter our approach to inflammatory processes and infectious diseases. The Satellite Symposium focused on recent advances in diversified fields including arachidonate metabolites, actions of cytokines, chemokines, cellular adhesion molecules and novel molecular approaches in the therapy of inflammation. Scholarly addresses were presented by eminent experts in their respective fields. In this book are included 17 papers which describe in detail novel approaches to the study of inflammation. We hope that this book will serve not only as a summary of present knowledge, but will stimulate both scientists and clinicians to continue their efforts to battle inflammatory diseases. W. Gene therapy for inflammatory diseases is a new , burgeoning field of medicine. Edited by the undisputed pioneers of this area of research, this volume is the first devoted to its topic. It contains thirteen chapters, each written by leaders in their respective fields, that summarize the state of the art in developing novel, gene based treatments for inflammatory diseases. As well as providing an introduction to the basic concepts of gene therapy and the use of naked DNA approaches, the book describes the advances that have been made in applying them to arthritis, lupus, multiple sclerosis, diabetes, Sjogren`s syndrome and transplantation. One chapter is devoted to discussing the first human clinical trials that apply gene therapy to the treatment of an inflammatory disease. As well as providing novel therapeutic approaches, gene therapy facilitates the development of new and improved animal models of disease; a chapter describing these advances is also included. As an up-to-date, timely book written by th

In November 1998 many of the key leaders of new drug discovery for inflammatory diseases gathered at Hershey, Pennsylvania for the 9th International Conference of the Inflammation Research Association. The Conference was held over a five day period and provided a stimulating environment for the open exchange of important advances in basic inflammation research as well as new drug discovery and development. This book encompasses some of the highlights of several presentations made at the Conference. It contains some of the latest and important developments in the field of inflammation research. Topics include the status of eotaxin and chemokines in asthma and allergy, signal transduction and regulation of diverse mediators such as the JNK group of MAP kinases, TNF and IL-1 signaling of NF-kB as well as regulators of AP-1, macrophage metalloproteinases, lymphotoxin and further insights into the role of MCP-1 in disease. Also discussed are drug targets in rheumatoid and osteoarthritis, fibrotic diseases,...

Improving and Accelerating Therapeutic Development for Nervous System Disorders is the summary of a workshop convened by the IOM Forum on Neuroscience and Nervous System Disorders to examine opportunities to accelerate early phases of drug development for nervous system drug discovery. Workshop participants discussed challenges in neuroscience research for enabling faster entry of potential treatments into first-in-human trials, explored how new and emerging tools and technologies may improve the efficiency of research, and considered mechanisms to facilitate a more effective and efficient development pipeline. There are several challenges to the current drug development pipeline for nervous system disorders. The fundamental etiology and pathophysiology of many nervous system disorders are unknown and the brain is inaccessible to study, making it difficult to develop accurate models. Patient heterogeneity is high, disease pathology can occur years to decades before becoming clinically apparent, and diagnostic and treatment biomarkers are lacking. In addition, the lack of validated targets, limitations related to the predictive validity of animal models - the extent to which the model predicts clinical efficacy - and regulatory barriers can also impede translation and drug development for nervous system disorders. Improving and Accelerating Therapeutic Development for Nervous System Disorders identifies avenues for moving directly from cellular models to human trials, minimizing the need for animal models to test efficacy, and discusses the potential benefits and risks of such an approach. This report is a timely discussion of opportunities to improve early drug development with a focus toward preclinical trials.
