This volume focuses on recent advances in the biochemical and molecular analysis of different families of phospholipases in plants and their roles in signaling plant growth, development and responses to abiotic and biotic cues. The hydrolysis of membrane lipids by phospholipases produces different classes of lipid mediators, including phosphatidic acid, diacylglycerol, lysophospholipids, free fatty acids and oxylipins. Phospholipases are grouped into different families and subfamilies according to their site of hydrolysis, substrate usage and sequence similarities. Activating one or more of these enzymes often constitutes an early, critical step in many regulatory processes, such as signal transduction, vesicular trafficking, secretion and cytoskeletal rearrangements. Lipid-based signaling plays pivotal roles in plant stress responses, cell size, shape, growth, apoptosis, proliferation, and reproduction.

Cell membranes are the initial and focal sites of stimulus perception and signal transduction. Membrane lipids are rich sources for the production of signaling messengers that mediate plant growth, development, and response to nutrient status and stresses. In recent years, substantial progress has been made toward understanding lipid signaling in plants, but many fundamental questions remain: What lipids are signaling messengers or mediators in plants? How are the signaling lipids produced and metabolized? In what plant cellular and physiological processes are various lipid mediators involved? How do they carry out their signaling functions? How do lipid signaling networks contribute to modulating plant growth, development, and responses to hormones and stresses? In this Research Topic issue, we invite the broad plant community to address the above questions.Cell membranes are the initial and focal sites of stimulus perception and signal transduction. A collection of papers that comprehensively describe the major areas of research on lipid metabolism of plants. State-of-the-art knowledge about research on fatty acid and glycerolipid biosynthesis, isoprenoid metabolism, membrane structure and organization, lipid oxidation and degradation, lipids as intracellular and extracellular messengers, lipids and environment, oil seeds and gene technology is reviewed. The different topics covered show that modern tools of plant cellular and molecular biology, as well as molecular genetics, have been recently used to characterize several key enzymes of plant lipid metabolism (in particular, desaturases, thioesterases, fatty acid synthetase) and to isolate corresponding cDNAs and genomic clones, allowing the use of genetic engineering methods to modify the composition of membranes or storage lipids. These findings open fascinating perspectives, both for establishing the roles of lipids in membrane function and intracellular signalling and for adapting the composition of seed oil to the industrial needs. This book will be a good reference source for research scientists, advanced students and industrialists wishing to follow the considerable progress made in recent years on plant lipid metabolism and to envision the new opportunities offered by genetic engineering for the development of novel oil seeds.

Phospholipases generate lipid signaling molecules through their hydrolytic action on phospholipids and are known to regulate function of a variety of cells under normal and diseased conditions. While several physiological, biochemical and molecular techniques have identified key players involved in different disease processes, phospholipases have also emerged as critical players in the pathogenesis of a number of different diseases including cancer and heart disease. In addition, phospholipases are also implicated in such conditions as brain disorder/injury, kidney and immune cell dysfunction. Phospholipases in Health and Disease is a compilation of review articles dedicated to the study of the field with respect to biochemical and molecular mechanisms of normal and abnormal cell function. The wide range of area covered here is of interest to basic research scientists, clinicians and graduate students, who are engaged in studying pathophysiological basis of a variety of diseases. Furthermore, this book highlights the potential of the different phospholipases as therapeutic targets as well as part of prevention strategies. Twenty three articles in this book are organized in four sections that are designed to emphasize the most characterized forms of the phospholipases in mammalian cells. The first section discusses general aspect of phospholipases. Section two covers the role and function of phospholipase A in different pathophysiological conditions. The third section is focussed on phospholipase C which is believed to play a central role in transmembrane signaling. The final section covers phospholipase D which is present in a variety of different cells. The book illustrates that the activation of phospholipases is of fundamental importance in signal transduction affecting cell function. Overall, this book discusses the diverse mechanisms of phospholipase mediated signal transduction in different pathophysiological conditions and raises the possibility of specific forms of phospholipases serving as novel targets for drug development.

The highly structured eucaryotic cell with its complex division of biochemical labour requires a distinct protein complement in each cellular structure and compartment. Nuclear coded and cytosolically synthesized polypeptides are specifically sorted to every corner of the cell in a post- or co-translational manner. The presence of separate genomes and protein translation machineries in plastids and mitochondria requires further coordination not only on the transcriptional, translational but also most likely on the protein import level. Numerous different protein transport systems have developed and coexist within plant cells to ensure the specific and selective composition of every sub-cellular compartment. This volume summarizes the current knowledge on protein trafficking in plant cells. Aside from the fundamental aspects in cell biology of how specific pre-protein sorting and translocation across biological membranes is achieved, a major focus is on transport, modification and deposition of plant storage proteins. The increasing use of plants as bioreactors to provide custom-designed proteins of different usage requires detailed understanding of these events. This text is directed not only at students and professionals in plant cell and molecular biology but also at those involved in horticulture and plant breeding. It is intended to serve as a text and guide for graduate-level courses on plant cell biology and as a valuable supplement to courses in plant physiology and development. Scientists in other disciplines who wish to learn more about protein translocation in plants will also find this text an up-to-date source of information and reference.

This book examines signal-activated phospholipases, a class of cellular enzymes activated in response to hormones, neurotransmitters, growth factors, cytokines, and other agonists. The signal-activated phospholipases produce a host of lipid-derived second messengers that mediate agonist and cell type-specific responses.
