Heterogeneity, or mixtures, are ubiquitous in genetics. Even for data as simple as mono-genic diseases, populations are a mixture of affected and unaffected individuals. Still, most statistical genetic association analyses, designed to map genes for diseases and other genetic traits, ignore this phenomenon. In this book, we document methods that incorporate heterogeneity into the design and analysis of genetic and genomic association data. Among the key qualities of our developed statistics is that they include mixture parameters as part of the statistic, a unique component for tests of association. A critical feature of this work is the inclusion of at least one heterogeneity parameter when performing statistical power and sample size calculations for tests of genetic association. It is true that modern teaching is faced with heterogeneous students. Ironically, this is not a recent development: students have always been different. Consequently, there is a broad discourse on "heterogeneity" in education. On the normative level of meta-narratives about modern democracy one will find the idea that more and more people have to be included in the modern welfare state. Nevertheless, before talking about inclusion one has to deal with the mechanisms of exclusion, if one is interested in the phenomenon of heterogeneity. At the heart of it, one will find the debate on the meaning of differences between students from an age group and their implications for school-based learning. Even more basically, Didaktik has to give a response on the dilemma arising of balancing "individual" and "collective" modes of teaching. However, Didaktik theory speaks in the singular and in the light of normality. It normally speaks of generalized homogenized students, axis-constructions and vertexes in the singular, denying their heterogeneity. How can the teacher's relation to this simultaneous heterogeneity be carried out in a justified way? The central idea of the book is to explore whether this professional Didaktik challenge can be studied through the concept of the others' "strangeness". This volume analyzes the constructions of heterogeneity in pedagogy based on the leitmotif of the stranger. In doing so, the stranger is seen as a didactic key. The book shows that there is a necessity to understand the filter of strangeness/otherness. Plasma cells (PCs) are terminally differentiated B-cells producing large amounts of immunoglobulins (Ig). In humans, most of circulating Ig are produced by bone marrow plasma cells. PCs differentiate from activated naïve or memory B-cells usually activated by specific antigens. It is still controversial whether the regulation of PCs numbers and the “active” in vivo Ig diversity depend or not on non-specific reactivation of B-cells during infections. Depending on the stimulus (T-independent/T-dependent antigen, cytokines, partner cells) and B-cell types (naïve or memory, circulating or germinal center, lymph nodes or spleen, B1 or B2...), both the phenotype and isotype of PCs differ suggesting that PC diversity is either linked to B-cell diversity or to the type of stimulus or to both. Knowledge of the mechanisms supporting PC diversity has important consequences for the management of i) plasma cell neoplasia such as Multiple Myeloma and Waldenström's Macroglobulinemia, ii) vaccine protection against pathogens and iii) auto-immune diseases.

Asthma is a chronic relapsing airways disease that represents a major public health problem worldwide. Intermittent exacerbations are provoked by airway mucosal exposure to pro-inflammatory stimuli, with RNA viral infections or inhaled allergens representing the two most common precipitants. In this setting, inducible signaling pathways the airway mucosa play a central role in the initiation of airway inflammation through production of antimicrobial peptides (defensins), cytokines, chemokines and arachidonic acid metabolites that coordinate the complex processes of vascular permeability, cellular recruitment, mucous hyper-secretion, bronchial constriction and tissue remodeling. These signals also are responsible for leukocytic infiltration into the submucosa, T helper-lymphocyte skewing, and allergic sensitization. Currently, it is well appreciated that asthma is a heterogeneous in terms of onset, exacerbants, severity, and treatment response. Current asthma classification methods are largely descriptive and focus on a single aspect or dimension of the disease. An active area of investigation on how to collect, use and visualize multidimensional profiling in asthma. This book will overview multidimensional profiling strategies and visualization approaches for phenotyping asthma.
