Biopolymers deals with the methods of physical characterization and the principles underlying them, with emphasis on quantitative aspects of sequence, conformation, and structure in both laboratory-synthesized and native biopolymers. The book reviews structural units of biopolymers and describes characterization of biopolymers, the available techniques, the evaluation of underlying principles, and experimental applications. Some of these methods include Raman spectroscopy, theoretical conformation analysis, electron microscopy, and morphology of laboratory-synthesized polymers. The text explains the factors controlling conformation of polypeptides, the steric maps of dipeptides, potential energy maps, and the calculation of tertiary polypeptide structure. The investigator can use X-ray diffraction to determine the structure of polymers and macromolecules, such as diffraction by a crystal, by poorly crystalline polymer systems, or by a helical chain. The book notes that materials that can be crystallized from strong solvents reveal morphology similar to that of commercial polymers, which are different from that of polypeptides or proteins in native tissue. The text explains the basis of infrared and Raman spectroscopy in probing molecular structure and conformation of biological macromolecules. The investigator can also employ nuclear magnetic resonance and dielectric relaxation for conformation in physical organic chemistry, outside of biological macromolecule applications. The book can prove helpful for researchers in ultra-trace analysis, polymer research, and analytical chemistry.

A thorough understanding of stereochemistry is essential for the comprehension of almost all aspects of modern organic chemistry. It is also of great significance in many biochemical and medicinal disciplines, since the stereoisomers of a compound can have dramatically different biological properties. This text explains how the different properties of stereoisomers of a compound arise, and what processes can be used to prepare and analyze stereoisomerically pure compounds. It also presents prominent coverage of the stereochemistry of inorganic and organometallic compounds, which is likely to increase in importance, as these compounds are used as symmetric catalysts in asymmetric synthesis. Modern stereochemical terminology is used throughout, although reference is also made to older terms which are still widely used. A set of problems at the end of each chapter aims to further the reader's understanding of how the content can be applied. The book is designed mainly as a textbook for undergraduate students and as a reference source for more advanced levels, but is also intended for academic and professional organic chemists.

The determination of the three-dimensional structure of a biological molecule is the starting point in the understanding of molecular mechanisms involved in its complex biochemical reactions. The molecular architecture of multimolecular systems such as membranes and chromosomes provides the key to the fascinating field of molecular biology. Stereochemical details of biological macromolecules and their interactions with pharmacological agents form the basis for drug design. Naturally, the study of the structure and function of biological molecules has aroused tremendous interest and investigations in this area are being carried out in a large number of laboratories. The techniques used for this purpose include both experimental methods (X-ray and neutron diffraction measurements, study of NMR, ESR, vibrational and electronic spectra, ORD, CD and dipole moment measurements, biochemical modifications etc. ) and the oretical methods (quantum mechanical and classical potential energy calculations, Monte Carlo simulations and molecular graphics). F or several years now, X-ray diffraction [1] has served as our only source of infor mation on the three-dimensional arrangements of atoms in biopolymers. Fiber-diffrac tion of DNA led to the proposal of the DNA double helix. Fibers of long~hain polymers show ordering in the direction of the fibre-axis but not in the transverse plane. Accurate estimates of the dimensions of helical structures can be made using techniques on the basis of which models of biopolymers can be constructed.

The prediction of the conformation of proteins has developed from an intellectual exercise into a serious practical endeavor that has great promise to yield new stable enzymes, products of pharmacological significance, and catalysts of great potential. With the application of predic tion gaining momentum in various fields, such as enzymology and immunology, it was deemed time that a volume be published to make available a thorough evaluation of present methods, for researchers in this field to expound fully the virtues of various algorithms, to open the field to a wider audience, and to offer the scientific public an opportunity to examine carefully its successes and failures. In this manner the practitioners of the art could better evaluate the tools and the output so that their expectations and applications could be more realistic. The editor has assembled chapters by many of the main contributors to this area and simultaneously placed their programs at three national resources so that they are readily available to those who wish to apply them to their personal interests. These algorithms, written by their originators, when utilized on pes or larger computers, can instantaneously take a primary amino acid sequence and produce a two-or three-dimensional artistic image that gives satisfaction to one's esthetic sensibilities and food for thought concerning the structure and function of proteins. It is in this spirit that this volume was envisaged.

The goal of this book is to give a systematic description of the main principles of affinity modification and applications, consideration of possibilities, and restrictions of the method. Modification within specific complexes is a special case of chemical modification which is widely used in the nonaddressed version in biochemistry and related areas. Therefore, we have included in the first introductory paper chapter of the book general considerations of chemical modifications of biopolymers and the application of biopolymers.
