Rapid advances in molecular biology have accelerated the production of a great number of protein-based therapeutic agents. The major cost in producing these proteins appears to be associated with their purification from the complex mixture of the crude extract. A major challenge to the protein biochemist and the biochemical engineer is the development of rapid, efficient, and cost-effective purification systems. This volume presents state-of-the-art reviews of current methods used in the purifica tion of biological macromolecules that are based on molecular interactions. Thus, the major emphasis is placed on affinity-related techniques. Part I provides a general introduction to affinity chromatography and includes a chapter describing an interesting new technique called "slalom chromatography" for DNA fractionations. Affinity chromatography using molecules of biological origin as the affinity ligand is covered in Part II. Part III describes the use of a special class of biomolecules, antibodies, as affinity ligands. Affinity chromatography with biomimetic ligands is discussed in Part IV. Newer concepts and their applications in bioseparation are presented in Part V. First published in 2004, this book collects several up-to-date methods for quantitative analysis of biospecific interactions, a field that has a long history that perhaps can be said to have begun with the classical paper of G. Scatchard in 1949 (The attractions of proteins for small molecules and ions, but which has advanced impressively during the last few years. A precise spatial arrangement of just a few hydrogen bonds can confer a remarkably specific reversible association between two molecules. A web of weak interactions governs biospecific recognition in general. The binding equilibria in living cells tune and coordinate a multitude of functions. The thermodynamic properties of such interactions are often studied by binding experiments in simplified and essentially ideal systems. However, similar types of studies may elucidate the biologically relevant dynamic steady-state conditions in living cells and organisms, allowing for the very wide range of interactant concentrations and the interplay between the many reactions and interactions. The development in biosciences will continue with in-depth studies of macromolecules and membranes. More detailed knowledge will allow analyses of delicate balances between substances and events in the complex systems involved in life processes. Methods to study biospecific affinities are thus highly important tools for understanding mechanisms and effects of molecular binding events in vivo and in vitro, e.g., in biochemical, biomedical and pharmaceutical research, and for biotechnological research and production.

A NATO Advanced Research Workshop entitled New Methods for the Study of Molecular Aggregates was held at Tbe Lodge at Kananaskis Village, Alberta, Canada from 16 -20 June 1996. In fact the meeting was entirely concerned with the problem of analyzing biomolecular complexes, so the title of these proceedings has been altered to give a more precise description of the content. Tbe workshop was hosted by the time-of-flight group of the Department of Physics at the University of Manitoba, and was attended by 64 participants from around the world. '!\venty-one invited talks were given and 27 papers were presented as posters. Of the 48 contributions, 22 papers (12 orals, 10 posters) are included in these proceedings. Tbe subject of the conference was the investigation of noncovalent biomolecular complexes, with particular focus on the application of mass spectrometry to their characterization. '!\vo new ionization techniques introduced in the late 1980s, electrospray ionization (ES I) and matrix-assisted laser desorptionlionization (MALDI), resulted in a breakthrough in mass spectrometry, enabling its use in molecular weight and primary structure determination of biopolymers larger than 100 kDa. Recently it has been discovered that ESI mass spectrometry mayaiso be used to characterize complexes containing noncovalent interactions, thus opening new perspectives for supramolecular chemistry. ESI mass spectrometry has the advantage that the sampie is introduced from a homogenous solution which can be maintained at near physiological conditions of pR, concentration, and temperature.
